Local Contact Startup Guidelines.

When this protocol is successful, users have the assurance that the beamline will give them data fitting with the quality assessment

Aim of this protocol: To help the LC startup the beamlines quickly and efficiently. Please use the log book to note current readings and anomalies.

Write down in the beamline logbook the following information :

  • Date
  • Experiment number (e.g. MX-415)
  • Your name
  • Your phone number on site and off-site, may also be written on white board.

Inside the experimental hutch :

  • Check that the hutch is clean (if not note this in the log book and mail to BLSci)
  • Check that all tools are in/on tool box/board
    • Gloves and glasses are available to handle liquid N2.
    • Glasses for laser protection.
  • Check that no sample is on the goniometer
  • Check that the cryo stream is aligned and working, level of N2 is ok,  temperature regulation @ 100K
  • Put the YAG screen on the goniometer (except ID23eh2)
  • Check that exes_hutch is working - use it to align the YAG screen.
  • Close the experimental hutch.

Align the beamline :

  • Check that all applications needed on the end-station are running :
    • Bliss device servers
    • Front end and Vacuum applications.
    • Spec oh
    • Spec eh
    • mxControlPanel (Id23-1,2 and Id29)
    • mxCuBE : log in as users
    • Firefox and ISPyB
    • restart DNA (after logging into mxCuBE)
    • Image viewer
  • Do a Realign of the beamline (except ID23-1 -> use LC_startup in eh1).
  • In the logbook write  (in addition ID23-1 -> use LC_startup in eh1) :
    • Machine status : ring current and operation mode.
    • Check Front end is open.
    • Check that automatic mode will last the experiment time.
    • The energy if tuneable beamline
    • Undulator Gaps
    • The values of the diodes along the beam path with all shutters open and 100% transmission and slits open to beamline standard sizes (note also the gain)
  • Check that these values fit the standards of the beamline (compare at least with values of the day before).
  • With the safety shutter and fast shutter open, check that beam is centred.

Verify data collection with user account:

  • Collect with mxCuBE at 1.7Å a blank image, should be free of beamstop artefacts.
  • If test Xtals use one to collect images with mxCuBE at 1.7Å
  • Same thing with DNA
  • Check that ISPyB has been updated.

Sample changer:

  • Check that the gonio head is the Smart Magnet head or miniKappa.
  • Check that there is no baskets in the sample changer.
  • If not tested before, put a test basket (with test samples) then test the sample changer (load/unload sample) using mxCuBE.

Safety and other stuff with users presence:

  • Check that users have done their safety training.
  • Explain about the oxygen level alarm and what to do if it sounds.
  • Check that users know how to close the experimental hutch door.
  • Check that users are confident with using the beamline.
  • Check that users know the door code.
  • Leave phone numbers where you can be contacted at all times (until 10 pm).

For ID23-2 only

 

  • Note in logbook pressure on the KB vessel  - check pressure sensor (blue device at the left side of the KB vessel) (985-1000 mbar).
  • Check that helium cylinder (in the optics hutch electronic rack).
